چکیده

With the availability of several completed genome sequences, the analyses of large-scale interactions among the proteins encoded by them become important to decipher the complexities of biological systems involving intricate networks of macromolecular interactions. Various approaches have been developed to generate systematic genome-wide protein interaction maps (PIMs), which provide functional information of many unknown proteins. Among many methods, the yeast two-hybrid (Y2H) system is popular because it is an in vivo and inexpensive technique and easily amenable for genome-wide application. Analyses of proteomes of E. coli virus T7, vaccinia virus, yeast Saccharomyces cerevisiae, human pathogen Helicobactor pylori and nematode Caenorhabditis elegans have been carried out using Y2H. The resulting PIMs have identified functions of many proteins and generated several new hypotheses for further experimentation.

Yeast Two Hybrid cDNA Screening of Arabidopsis thaliana for SETH4 Protein Interaction

SETH4 coding sequence with 2013 bp is a member of gene family expressed in gametophytic tissues of Arabidopsis thaliana. This fragment was PCR amplified using Kod Hi Fi DNA polymerase enzyme. This fragment was cloned into pGBKT7 bate vector and transformed E. coli DH5? cells containing vector were selected on LB medium containing Kanamycin. Finally, pGBKT7-SETH4 bate was transformed into yeast ...

A strategy for constructing large protein interaction maps using the yeast two-hybrid system: regulated expression arrays and two-phase mating.

Maps representing the binary interactions among proteins have become valuable tools for understanding how proteins work together to mediate biological processes. One of the most effective methods for detecting biologically important protein interactions has been the yeast two-hybrid system. Here we present an efficient two-hybrid strategy to facilitate construction of protein interaction maps o...

Protein--protein interaction maps: a lead towards cellular functions.

The availability of complete genome sequences now permits the development of tools for functional biology on a proteomic scale. Several experimental approaches or in silico algorithms aim at clustering proteins into networks with biological significance. Among those, the yeast two-hybrid system is the technology of choice to detect protein-protein interactions. Recently, optimized versions were...

A densely overlapping gene fragmentation approach improves yeast two-hybrid screens for Plasmodium falciparum proteins.

Use of the yeast two-hybrid assay to study Plasmodium falciparum protein-protein interactions is limited by poor expression of P. falciparum genes in yeast and lack of easily implemented assays to confirm the results. We report here two methods to create gene fragments - random fragmentation by partial DNAse I digestion and generation of densely overlapping fragments by PCR - that enable most p...

Bayesian Inference of Protein and Domain Interactions Using The Sum-Product Algorithm

In order to fully understand the functions of proteins in living organisms we must study their interactions and construct accurate interaction maps. Each protein can be composed of one or several peptide chains called domains and each protein interaction can be seen as a consequence of an underlying interaction of two domains, one from each protein. Since high-throughput methods of measuring pr...
